Overview

Ivonescimab, known as SMT112, is a novel, potential first-in-class investigational bispecific antibody that combines immunotherapy (PD-1 blockade) with anti-angiogenesis (VEGF blockade) into a single molecule. Summit has begun its clinical development of ivonescimab in NSCLC, with three active Phase III trials: HARMONi, HARMONi-3, and HARMONi-7. HARMONi is a Phase III trial evaluating ivonescimab with chemotherapy vs placebo plus chemotherapy in EGFR-mutated locally advanced or metastatic non-squamous NSCLC after progression on a 3rd generation EGFR TKI (e.g., osimertinib). HARMONi-3 is a Phase III trial evaluating ivonescimab with chemotherapy vs pembrolizumab with chemotherapy in first-line metastatic NSCLC. HARMONi-7 is a Phase III trial evaluating ivonescimab monotherapy vs pembrolizumab monotherapy in first-line metastatic NSCLC with high PD-L1 expression. Ivonescimab is investigational and not approved by regulatory authorities in Summit’s license territories (including the U.S. and Europe). It was approved for marketing authorization in China in May 2024 and has received Fast Track designation by the U.S. FDA for the HARMONi setting.
