Purpose We are seeking an accomplished Sr. Scientist with deep expertise in higher order structure (HOS) characterization of Antibody-Drug Conjugates (ADCs) to join our Biophysics team. The ideal candidate will play a pivotal role in supporting the structural integrity assessment of ADCs in drug discovery. The Scientist will lead the development and application of advanced biophysical and structural methodologies to understand conjugation effects on antibody structure, stability, and function.
You Will Be Responsible To
Serve as a HOS subject matter expert (SME) of ADCs, designing and performing structural studies to assess: 1) impact of drug conjugation on antibody folding, domain structure, and inter-chain interactions; 2) conformational changes, aggregation propensity, and stability profiles.
Develop, optimize, and interpret data from advanced HOS analytical techniques.
Lead training initiatives on HOS techniques and ADC-specific structural characterization challenges, including forced degradation studies and linker stability.
Continuously evaluate new biophysical technologies and workflows for high-resolution structural assessment of complex ADC molecules and new modalities.
Apply the workflows to study antibody conformational stability, domain interactions, and aggregation mechanisms under formulation and stress conditions.
Collaborate with cross-functional and multidisciplinary teams in Formulation Development, Protein Engineering, Data Science, MS characterization and Binding group.
Analyze complex datasets using advanced analytical and computational tools.
Present research findings at internal and external scientific meetings and symposiums.
Qualifications
Education:
M.Sc with 5+ years of work experience or Ph.D. and 0‑3+ years of work experience in Biophysics or Biochemistry, Structural biology, Analytical chemistry, or a closely related field.
Required Experience and Skills:
Proven experience with HOS characterization of ADCs, including an understanding of drug-to-antibody ratio (DAR), stability, and structural heterogeneity.
Experience with biophysical techniques (DSC, DLS, DSF, HPLC) and strong background in spectroscopy (fluorescence, UV‑Vis, Infrared, and NMR), light scattering, calorimetry, and computational modeling.
Excellent problem-solving skills and a strong publication record.
Preferred: Experience with data analysis software, programming languages (Python, AI/ML-based programming), and simulation tools (PyMOL, MOE, Schrodinger, Rosetta, etc.).
Basic knowledge of kinetics and thermodynamics properties and their applications in characterizing protein structure and dynamics.
