Summit Therapeutics, Inc.
About Summit
Ivonescimab, known as SMT112, is a novel, potential first‑in‑class investigational bispecific antibody that combines the effects of immunotherapy via PD‑1 blockade with anti‑angiogenesis through VEGF inhibition in a single molecule. It displays unique cooperative binding to each target with higher affinity when both PD‑1 and VEGF are present. Summit has begun clinical development of ivonescimab in non‑small‑cell lung cancer (NSCLC) with three active Phase III trials:
HARMONi – evaluate ivonescimab plus chemotherapy versus placebo plus chemotherapy in patients with EGFR‑mutated, locally advanced or metastatic non‑squamous NSCLC who progressed after a third‑generation EGFR TKI.
HARMONi‑3 – evaluate ivonescimab plus chemotherapy versus pembrolizumab plus chemotherapy in patients with first‑line metastatic NSCLC.
HARMONi‑7 – evaluate ivonescimab monotherapy versus pembrolizumab monotherapy in patients with first‑line metastatic NSCLC and high PD‑L1 expression.
Ivonescimab is investigational and not yet approved in most Summit license territories, including the United States and Europe. It was approved for marketing authorization in China in May 2024 and received Fast Track designation by the FDA for the HARMONi clinical trial setting.
